Drug intelligence / Profile preview
EO4010 + nivolumab + bevacizumab is an investigational combination regimen consisting of Enterome's synthetic peptide-based therapeutic cancer vaccine EO4010, Bristol Myers Squibb's anti-PD-1 checkpoint inhibitor nivolumab, and Genentech/Roche's anti-VEGF-A monoclonal antibody bevacizumab. EO4010 is designed to expand pre-existing memory CD8+ T cells that recognize non-self gut bacterial peptides cross-reacting with five tumor-associated antigens (TAAs) upregulated in colorectal cancer: BIRC5 (survivin), FOXM1, UBE2C, CDC20, and KIF2C. It also includes the CD4+ helper peptide UCP2 (derived from hTERT) to enhance immune activation. The combination was evaluated in the Phase 1/2 AUDREY trial (NCT05589597) for patients with previously treated, unresectable, microsatellite-stable (MSS) and mismatch repair proficient (pMMR) metastatic colorectal cancer. Although the trial demonstrated safety, immunogenicity, and preliminary efficacy, it was terminated by the sponsor.
