Overview

This investigational combination therapy consists of intraperitoneally administered CAR-T cells targeting the Epithelial Cell Adhesion Molecule (EpCAM) and a lymphodepleting preconditioning chemotherapy regimen of cyclophosphamide and fludarabine. Developed by Jian-Kun Hu at West China Hospital, Sichuan University, the treatment is specifically designed for patients with advanced gastric cancer and peritoneal metastasis. The CAR-T cells are genetically engineered to recognize and eliminate EpCAM-expressing tumor cells, which are prevalent in gastric malignancies. The chemotherapy component (cyclophosphamide 300 mg/m² and fludarabine 30 mg/m²) is administered prior to the CAR-T cell infusion to deplete host lymphocytes, thereby creating a more favorable environment for the expansion, persistence, and anti-tumor activity of the infused cell therapy.
