"epcam-sn-38 + nucleoside analogs + rnai" is a multi-payload RNA nanoparticle therapeutic being developed by RNA NanoBiotics for the treatment of metastatic colorectal cancer (mCRC). The platform utilizes an RNA-based nanoparticle functionalized to target the Epithelial Cell Adhesion Molecule (EpCAM), a protein frequently overexpressed in colorectal tumors. The nanoparticle co-delivers three distinct therapeutic agents: SN-38 (the active metabolite of the topoisomerase I inhibitor irinotecan), nucleoside analogs (which interfere with DNA synthesis), and RNA interference (RNAi) molecules designed to silence specific oncogenic pathways. This combinatorial approach aims to achieve synergistic anti-tumor effects while minimizing systemic toxicity through targeted delivery. The program is currently in preclinical development.
