Overview

**Epirubicin + carboplatin + gemcitabine** is a triplet chemotherapy combination used investigatively in advanced cancers. **Epirubicin**, an anthracycline antibiotic, intercalates DNA and inhibits topoisomerase II, causing DNA damage and cell death. **Carboplatin**, a platinum-based alkylating agent, forms DNA crosslinks that block replication and transcription. **Gemcitabine**, a nucleoside analog, incorporates into DNA during synthesis, leading to chain termination and inhibition of ribonucleotide reductase. This regimen is evaluated in phase II trials for cancers including breast cancer, ovarian cancer, hepatocellular carcinoma, and non-small cell lung cancer, showing response rates of 20-70% depending on indication, with common toxicities including grade 3-4 neutropenia, thrombocytopenia, and anemia; it is administered intravenously every 3 weeks.
