Overview

EOF5 is a combination chemotherapy regimen consisting of epirubicin, oxaliplatin, and a 5-day continuous infusion of 5-fluorouracil. It is primarily investigated for the treatment of advanced or metastatic gastric cancer. The regimen combines three distinct antineoplastic agents to inhibit tumor growth through multiple pathways: epirubicin, an anthracycline that intercalates DNA and inhibits topoisomerase II; oxaliplatin, a platinum-based agent that forms DNA cross-links; and 5-fluorouracil, a pyrimidine analog that inhibits thymidylate synthase. The specific 5-day continuous infusion schedule of 5-fluorouracil is intended to maximize exposure during the S-phase of the cell cycle, potentially improving efficacy and tolerability compared to bolus administration. This regimen has been evaluated in Phase II clinical trials, particularly by Fudan University, for patients with unresectable advanced or metastatic gastric cancer.
