Epirubicin + tamoxifen is a combination regimen used primarily in the treatment of breast cancer, particularly in postmenopausal women with operable or advanced disease. Epirubicin is an anthracycline antibiotic that acts as a topoisomerase II inhibitor, interfering with DNA replication and transcription by intercalating into DNA and stabilizing the DNA-topoisomerase II complex. Tamoxifen is a selective estrogen receptor modulator (SERM) that antagonizes estrogen receptors on breast cancer cells, thereby inhibiting tumor growth driven by estrogen signaling. The combination has demonstrated synergistic effects, including enhanced anti-tumor activity and suppression of angiogenic pathways such as VEGF/VEGFR2 signaling. This regimen may be used as adjuvant or neoadjuvant therapy to improve outcomes in hormone receptor-positive breast cancer[1][2][5].
