Overview

Epirubicin + valproic acid is an investigational combination therapy evaluated primarily in patients with solid tumor malignancies, including breast cancer. Epirubicin is an anthracycline antineoplastic agent that intercalates DNA and inhibits topoisomerase II, leading to inhibition of DNA replication and cell death. Valproic acid, traditionally used as an anticonvulsant, acts as a histone deacetylase (HDAC) inhibitor in this context, promoting histone acetylation and altering gene expression to induce differentiation and apoptosis of tumor cells. The combination aims to exploit the synergistic effects of HDAC inhibition by valproic acid with the cytotoxic action of epirubicin. Clinical studies have shown notable antitumor activity with acceptable toxicity profiles in heavily pretreated patients[1][2][7].
