ERAS-0015 + panitumumab is a combination therapy under clinical investigation for advanced or metastatic solid tumors, especially in patients with RAS-mutant cancers. **ERAS-0015** (also known as JYP-0015) is an orally bioavailable small molecule pan-RAS inhibitor, distinct as a molecular glue that targets all three major RAS isoforms (KRAS, NRAS, HRAS) by promoting formation of a ternary complex between active RAS and Cyclophilin A, thereby blocking downstream RAS/MAPK pathway signaling critical to tumor growth[2][4][8]. **Panitumumab** is a fully human monoclonal antibody targeting the epidermal growth factor receptor (**EGFR**), acting as a competitive inhibitor at the ligand-binding domain and inhibiting receptor activation and downstream signaling. The combination is intended to overcome resistance mechanisms seen in RAS-mutant tumors by targeting both RAS and EGFR pathways. ERAS-0015 is developed by Erasca (originally by Guangzhou JOYO Pharma/Joyo Pharmatech). Panitumumab is marketed under the brand name Vectibix and was developed and manufactured by Amgen[1][4][2][5][8].
