Drug intelligence / Profile preview
**ERAS-0015 + pembrolizumab** is an investigational combination regimen composed of ERAS-0015, an orally bioavailable pan-RAS molecular glue inhibitor, and pembrolizumab, a PD-1 immune checkpoint inhibitor monoclonal antibody (marketed as Keytruda). ERAS-0015 is designed to inhibit pathogenic RAS signaling in RAS-mutant and wildtype tumors, acting as a molecular glue to induce a complex between RAS and cyclophilin A, thereby disrupting oncogenic signaling in the RAS/MAPK pathway. Pembrolizumab is a humanized monoclonal antibody targeting the programmed cell death protein 1 (PD-1), restoring T-cell anti-tumor activity. The combination is being investigated in early-phase clinical trials for patients with advanced or metastatic solid tumors harboring RAS mutations, with the goal of addressing resistance mechanisms and improving anti-tumor responses in RAS-driven cancers[1][2][5][7].
