**ERAS-007 + encorafenib + cetuximab** is a combination therapy under investigation for the treatment of metastatic colorectal cancer (CRC) harboring the BRAF V600E mutation. ERAS-007 is a novel, orally bioavailable, selective inhibitor of ERK1/2, targeting a key component of the RAS/MAPK signaling pathway, which is frequently dysregulated in cancer. Encorafenib is a selective BRAF inhibitor, and cetuximab is a monoclonal antibody targeting the epidermal growth factor receptor (EGFR). The rationale for combining these agents is to block the RAS/MAPK pathway at multiple nodes: upstream at EGFR (cetuximab), midstream at BRAF (encorafenib), and downstream at ERK (ERAS-007), in an effort to overcome resistance mechanisms and improve efficacy for patients with BRAF V600E-mutant metastatic CRC. Early-phase clinical trials (HERKULES-3) show preliminary safety and encouraging clinical activity for this combination in both treatment-naïve and previously treated patients.
