Overview

Eravacycline + aminoglycosides is a combination antibacterial therapy consisting of eravacycline, a synthetic fluorocycline antibiotic of the tetracycline class, and an aminoglycoside antibiotic (such as amikacin). Eravacycline acts by binding to the 30S ribosomal subunit in bacteria, inhibiting protein synthesis and thereby exerting bacteriostatic or bactericidal effects depending on the organism. Aminoglycosides act primarily by binding to the bacterial 30S ribosomal subunit as well but cause misreading of mRNA, leading to defective protein synthesis and bacterial cell death. This combination has shown promising synergistic activity against multidrug-resistant Gram-negative pathogens such as carbapenem-resistant Acinetobacter baumannii (CRAB) and extensively drug-resistant strains. The rationale for combining these agents is to enhance antibacterial efficacy and potentially suppress resistance development in difficult-to-treat infections[1][3][10].
