ERC1671 + bevacizumab is a combination therapy under investigation for the treatment of recurrent glioblastoma multiforme (GBM) and gliosarcoma. ERC1671, also known as Gliovac, is an allogeneic/autologous therapeutic vaccine composed of whole, inactivated tumor cells and tumor cell lysates derived from both the patient and three GBM donors. The vaccine aims to stimulate a broad immune response against tumor antigens by presenting both autologous and allogeneic material to the patient's immune system. Bevacizumab is a monoclonal antibody that targets vascular endothelial growth factor A (VEGF-A), inhibiting angiogenesis within tumors. In clinical studies, this combination has been administered alongside granulocyte-macrophage colony-stimulating factor (GM-CSF) and cyclophosphamide to enhance immunogenicity. The addition of ERC1671/GM-CSF/cyclophosphamide to standard-of-care bevacizumab has shown improved overall survival compared with bevacizumab alone in patients with recurrent GBM[1][4][5]. The mechanism involves direct anti-tumor immunity via vaccination plus inhibition of tumor blood vessel formation by VEGF blockade.
