Overview
This is a **combination therapy** consisting of **erdafitinib**, a pan-fibroblast growth factor receptor (FGFR) kinase inhibitor; **cetrelimab**, a monoclonal antibody targeting programmed cell death protein 1 (PD-1); and **cisplatin**, a platinum-based chemotherapy agent. This regimen is being investigated for treating **locally advanced or metastatic urothelial carcinoma**, particularly in patients with FGFR genetic alterations and ineligibility for cisplatin-based therapy. Erdafitinib blocks FGFR1-4 and multiple other kinases, inhibiting tumor cell proliferation and survival. Cetrelimab blocks PD-1, promoting T-cell-mediated immune responses against cancer cells. Cisplatin induces cytotoxic DNA damage in cancer cells. Clinical trials, including the NORSE study, are evaluating safety, tolerability, and efficacy of this triple combination[3][7].
