A combination therapy consisting of ergoloid mesylates and 5-hydroxytryptophan (5-HTP) currently under investigation for the treatment of Fragile X syndrome. Ergoloid mesylates is a mixture of dihydrogenated ergot alkaloids (dihydroergocornine, dihydroergocristine, and dihydroergocryptine) that acts as a multi-receptor modulator with activity at dopaminergic, serotonergic, and adrenergic receptors. 5-HTP is a naturally occurring amino acid and metabolic precursor to the neurotransmitter serotonin. This specific combination was identified using artificial intelligence by the biotech company Purposeful and is being evaluated in a Phase 2 clinical trial led by Dr. Elizabeth Berry-Kravis at Rush University Medical Center. Preclinical studies in mouse models of Fragile X syndrome suggested that the combination could reverse behavioral and cognitive symptoms by synergistically modulating central neurotransmission.
