Eribulin + everolimus is an investigational combination therapy consisting of two small molecule drugs, eribulin and everolimus. Eribulin is a non-taxane microtubule dynamics inhibitor that disrupts mitotic spindle formation, leading to cell cycle arrest and apoptosis. Everolimus is an oral mTOR (mechanistic target of rapamycin) inhibitor that blocks the PI3K/AKT/mTOR signaling pathway, which is frequently dysregulated in various cancers including triple-negative breast cancer (TNBC). Preclinical studies have shown synergistic antitumor effects when these agents are combined, particularly in TNBC models. The combination has been evaluated in phase I clinical trials for safety and tolerability in patients with metastatic TNBC who have received prior chemotherapy[1][3][6]. Early results suggest modest efficacy with manageable toxicity profiles.
