Overview

**Eribulin + irinotecan** is an investigational combination therapy involving the use of eribulin, a synthetic microtubule dynamics inhibitor, and irinotecan, a topoisomerase I inhibitor. Eribulin (marketed as Halaven) disrupts microtubule polymerization, leading to mitotic blockade and apoptosis, while irinotecan (marketed as Camptosar) inhibits topoisomerase I, resulting in DNA damage and cell death. Preclinical and clinical studies have explored this combination for relapsed or refractory solid tumors, particularly pediatric and adolescent sarcomas such as Ewing sarcoma, rhabdomyosarcoma, and non-rhabdomyosarcoma soft tissue sarcoma. The rationale for combining these agents is their distinct, generally non-overlapping toxicity profiles and the potential for synergistic cytotoxicity. Both intravenous (for eribulin) and oral/intravenous (for irinotecan) administration schedules have been investigated[1][2][3][4].
