A combination therapy consisting of eribulin, a microtubule inhibitor, and mifepristone, a glucocorticoid receptor (GR) antagonist. This novel combination is designed to improve antitumor activity in metastatic triple-negative breast cancer (TNBC) and other advanced solid tumors. Eribulin inhibits the growth phase of microtubules and sequesters tubulin into nonproductive aggregates, leading to G2/M cell-cycle block, disruption of mitotic spindles, and ultimately apoptotic cell death. Mifepristone acts as an antagonist of the glucocorticoid receptor, potentially enhancing the antitumor effects of eribulin.
