Overview

Eribulin + tislelizumab is an investigational combination therapy consisting of eribulin, a non-taxane microtubule dynamics inhibitor, and tislelizumab, a humanized IgG4 monoclonal antibody targeting the programmed cell death protein 1 (PD-1). Eribulin inhibits microtubule growth leading to G2/M cell cycle arrest and apoptosis in cancer cells. Tislelizumab blocks PD-1 on immune cells, preventing its interaction with PD-L1/PD-L2 and thereby enhancing anti-tumor immune responses. This combination is being studied primarily for metastatic triple-negative breast cancer (mTNBC), where preclinical rationale suggests that eribulin may modulate the tumor microenvironment to enhance the efficacy of immune checkpoint inhibitors like tislelizumab[3]. Early clinical evidence indicates promising activity in heavily pretreated mTNBC patients[3][2][4].
