Eribulin + trastuzumab is a combination therapy used primarily in the treatment of HER2-positive advanced or metastatic breast cancer. Eribulin is a non-taxane microtubule dynamics inhibitor derived from halichondrin B, which inhibits tubulin polymerization, leading to cell cycle arrest at the G2/M phase and subsequent apoptosis. Trastuzumab is a recombinant humanized IgG1 monoclonal antibody that targets the extracellular domain of the human epidermal growth factor receptor 2 (HER2), blocking downstream signaling pathways involved in cell proliferation and survival, and mediating antibody-dependent cellular cytotoxicity. This combination has been studied as first-line therapy for HER2-positive metastatic breast cancer and as salvage therapy after failure of other regimens such as taxanes or T-DM1. Clinical trials have shown promising efficacy with manageable toxicity profiles[4][8][9].
