The combination of erlotinib, gefitinib, and metformin is not a standard or approved drug product but has been investigated in research settings for the treatment of non-small cell lung cancer (NSCLC), particularly in patients with activating EGFR mutations. Erlotinib and gefitinib are both epidermal growth factor receptor tyrosine kinase inhibitors (EGFR-TKIs) that target the EGFR pathway to inhibit tumor cell proliferation. Metformin is an oral biguanide antihyperglycemic agent commonly used for type 2 diabetes mellitus but has demonstrated antitumor properties in various cancers. Preclinical and clinical studies suggest that combining EGFR-TKIs with metformin may enhance efficacy by overcoming primary resistance to TKIs through mechanisms such as suppression of IGF-1R signaling or reversal of epithelial-mesenchymal transition[1][2][3].
