Erlotinib + gemcitabine + nab-paclitaxel is a combination regimen used primarily in the investigational treatment of advanced pancreatic ductal adenocarcinoma. Erlotinib is a small molecule tyrosine kinase inhibitor targeting the epidermal growth factor receptor (EGFR). Gemcitabine is a nucleoside analog that inhibits DNA synthesis, and nab-paclitaxel is an albumin-bound formulation of paclitaxel, a microtubule inhibitor that disrupts cell division. The combination seeks to maximize antineoplastic efficacy through complementary mechanisms: EGFR pathway inhibition (erlotinib), cytotoxic DNA synthesis inhibition (gemcitabine), and microtubule disruption (nab-paclitaxel). All three drugs are approved for the treatment of pancreatic cancer individually or in dual combinations, but the triple combination is under clinical investigation for improved outcomes, especially in basal-like subtypes of pancreatic cancer[1][2][3][4][5].
