Erlotinib + nab-paclitaxel is a combination regimen of two anticancer agents used primarily in the treatment of advanced solid tumors. Erlotinib is an oral small molecule inhibitor targeting the epidermal growth factor receptor (EGFR) tyrosine kinase, thereby blocking downstream signaling pathways involved in cell proliferation and survival. Nab-paclitaxel is a nanoparticle albumin-bound formulation of paclitaxel that disrupts microtubule function by stabilizing tubulin polymers, leading to mitotic arrest and apoptosis. The albumin-bound formulation enhances tumor delivery via gp60-mediated transcytosis and interaction with SPARC protein in the tumor stroma[6][8][10]. This combination has been studied for synergistic effects in cancers such as non-small-cell lung cancer (NSCLC) and pancreatic ductal adenocarcinoma—especially basal-like subtypes—where EGFR inhibition may provide additional benefit over chemotherapy alone[2][3][4]. The regimen is under investigation for improved efficacy compared to standard therapies but is not FDA-approved as a fixed combination; each component has individual approvals.
