Erlotinib + nimotuzumab is a dual targeting therapy of the epidermal growth factor receptor (EGFR). This combination therapy consists of erlotinib, a first-generation EGFR tyrosine kinase inhibitor (TKI), and nimotuzumab, a humanized IgG1 monoclonal antibody generated against the extracellular domain of EGFR. The combination has shown promising results in treating non-small cell lung cancer (NSCLC) with leptomeningeal metastases (LM), providing good tolerance and efficacy. Nimotuzumab binds to the extracellular domain of EGFR while erlotinib inhibits the intracellular tyrosine kinase domain, creating a dual blockade of EGFR signaling.
