The combination of erlotinib and pembrolizumab is a therapeutic approach that combines two distinct drugs with different mechanisms of action for the treatment of non-small cell lung cancer (NSCLC). Erlotinib is an epidermal growth factor receptor (EGFR) tyrosine kinase inhibitor that works by blocking the EGFR, a protein on cells that encourages cancer cell growth. It inhibits the intracellular phosphorylation of tyrosine kinase associated with EGFR. Pembrolizumab is a monoclonal antibody that binds to the programmed death-1 (PD-1) receptor to block interaction with its ligands, PD-L1 and PD-L2. This binding activates T-cell–mediated immune responses against tumor cells.
