Overview

Erlotinib + rapamycin is a combination therapy consisting of two small molecule inhibitors used primarily in oncology research. Erlotinib is an epidermal growth factor receptor (EGFR) tyrosine kinase inhibitor, while rapamycin (also known as sirolimus) is an inhibitor of the mammalian target of rapamycin (mTOR). The combination has been investigated for its potential to enhance antitumor activity and overcome resistance mechanisms in various cancers, including pediatric low-grade gliomas and non-small cell lung cancer. Preclinical and early clinical studies suggest that this regimen can be well tolerated, with some evidence of disease stabilization or response in select patient populations[1][2][3][4][5].
