Erlotinib + seribantumab is a combination therapy evaluated primarily in advanced non-small cell lung cancer (NSCLC), especially in patients with EGFR wild-type tumors. Erlotinib is a small molecule tyrosine kinase inhibitor targeting the epidermal growth factor receptor (EGFR), while seribantumab (MM-121) is a fully human IgG2 monoclonal antibody that binds to human epidermal growth factor receptor 3 (HER3/ErbB3). Seribantumab blocks heregulin (HRG/NRG)-mediated ErbB3 signaling and induces receptor downregulation. The combination was studied for its potential to improve outcomes by dual inhibition of EGFR and HER3 pathways, particularly in biomarker-selected populations with detectable HRG mRNA expression[1][2][5].
