Erlotinib + temsirolimus is a combination therapy that targets two different cellular pathways. Erlotinib is an epidermal growth factor receptor (EGFR) inhibitor, while temsirolimus is a mammalian target of rapamycin (mTOR) inhibitor. This combination was developed to overcome resistance to mTOR inhibitors, which is partially mediated by activation of EGFR. The combination has been studied in phase I clinical trials for patients with refractory solid tumors.
