The combination of erlotinib and vinorelbine has been studied in clinical trials for the treatment of non-small cell lung cancer (NSCLC). This combination brings together two distinct mechanisms of action to target cancer cells. Erlotinib works by blocking the epidermal growth factor receptor (EGFR), a protein found on the surface of both normal and cancer cells that encourages cell growth. By inhibiting EGFR, erlotinib can slow or stop cancer growth in tumors that have EGFR mutations or overexpression. Vinorelbine is a semi-synthetic vinca alkaloid that functions as a mitotic inhibitor. It disrupts microtubule assembly during cell division, specifically during metaphase, preventing cancer cells from dividing properly. Additionally, vinorelbine promotes apoptosis (programmed cell death) in cancer cells. This combination has been studied in Phase I clinical trials using both conventional and metronomic dosing schedules.
