Overview
Ervogastat + resmetirom is an oral combination therapy being developed by Madrigal Pharmaceuticals in collaboration with Pfizer for the treatment of metabolic dysfunction-associated steatohepatitis (MASH). The combination utilizes two distinct mechanisms of action to address hepatic lipid metabolism: ervogastat is a small molecule inhibitor of diacylglycerol O-acyltransferase 2 (DGAT-2), which catalyzes the final step of triglyceride synthesis, while resmetirom is a selective agonist of the thyroid hormone receptor-beta (THR-β), which increases hepatic fat metabolism and improves cholesterol profiles. By targeting both the synthesis and the clearance of hepatic lipids, the combination aims to provide synergistic effects in reducing liver fat and resolving MASH pathology. It is currently in Phase 1 clinical development.
