Eryaspase + polychemotherapy is a **combination drug regimen** comprising eryaspase, an L-asparaginase enzyme encapsulated in donor-derived red blood cells, administered together with polychemotherapy backbones for the treatment of advanced pancreatic cancer in the second-line setting. Eryaspase depletes circulating asparagine and glutamine by hydrolysis within red blood cells, selectively targeting cancer cells dependent on extracellular asparagine and glutamine for growth and survival[1][4][7][5]. In clinical trials, polychemotherapy included gemcitabine plus nab-paclitaxel, or irinotecan-based regimens such as FOLFIRI (5-fluorouracil, leucovorin, irinotecan). Developed primarily for patients with advanced pancreatic adenocarcinoma whose disease progressed after first-line systemic therapy, this regimen did not significantly improve overall survival compared with chemotherapy alone, but demonstrated improved disease control and a manageable safety profile. Eryaspase is under development by ERYTECH for additional indications including acute lymphoblastic leukemia[1][5][7].
