Overview

**Erythromycin + zinc acetate** is a topical combination formulation (typically 40 mg/ml erythromycin and 12 mg/ml zinc acetate) used primarily for treating acne vulgaris, including inflammatory lesions like papules and pustules. Erythromycin, a macrolide antibiotic, inhibits bacterial protein synthesis by binding to the 50S ribosomal subunit, effectively targeting *Propionibacterium acnes* and reducing inflammation; zinc acetate enhances skin healing, provides astringent and antiseptic effects, promotes metallothionein production to regulate copper, and improves antibiotic penetration while exhibiting bacteriostatic activity. Applied twice daily as a solution or gel mixed from powder and solvent, it shows efficacy comparable to oral tetracycline in clinical trials, with significant reductions in acne severity and lesion counts within 8-12 weeks, and is formulated for cutaneous use with a shelf life of 8 weeks post-mixing.[1][2][3][4][5][7][9]
