esat6-cfp10 + tuberculin purified protein derivative is a combination of two diagnostic reagents used for the detection of Mycobacterium tuberculosis infection. Tuberculin purified protein derivative (PPD) is a mixture of proteins derived from M. tuberculosis, traditionally used in the Mantoux skin test to detect latent or active tuberculosis infection by eliciting a delayed-type hypersensitivity (DTH) response[1][3]. ESAT6-CFP10 is a recombinant fusion protein composed of two highly immunogenic antigens, Early Secreted Antigenic Target 6 kDa (ESAT-6) and Culture Filtrate Protein 10 kDa (CFP10), which are absent in BCG vaccine strains and most non-tuberculosis mycobacteria[2][3]. This combination aims to improve specificity and sensitivity for TB diagnosis compared to PPD alone, as ESAT6-CFP10 can distinguish M. tuberculosis infection from BCG vaccination and environmental mycobacterial exposure[8][3]. Both components act as skin test reagents that stimulate T-cell mediated immune responses at the site of injection; their combined use may enhance diagnostic accuracy for TB.
