REcN-F/Ca is a synthetically bioengineered probiotic consortium developed for the treatment of type 2 diabetes mellitus (T2DM) and obesity. It consists of a reactive oxygen species (ROS)-tolerant strain of *Escherichia coli* Nissle 1917 (EcN), which has been iteratively selected for hydrogen peroxide resistance, and subsequently functionalized with a composite of fructooligosaccharides (FOS) and calcium carbonate (CaCO3). This formulation is designed to survive the harsh environment of the gastrointestinal tract and deliver therapeutic effects by reshaping the gut microbiota, specifically enriching butyrate-producing taxa such as Lachnospiraceae and Blautia. The resulting increase in short-chain fatty acids (SCFAs) and the activation of Peroxisome Proliferator-Activated Receptor (PPAR) signaling pathways lead to improved lipid metabolism, reduced adipose inflammation, and restored glucose homeostasis. In preclinical murine models, REcN-F/Ca has demonstrated significant efficacy in reducing weight gain, improving insulin sensitivity, and reversing metabolic dysfunction.
