Esculin and digoxin (often referred to as esculin and digitalis glycosides) is a compound ophthalmic preparation used primarily for the treatment of visual fatigue (asthenopia) and dry eye disease. The formulation combines **esculin**, a coumarin glycoside derived from horse chestnut (*Aesculus hippocastanum*), with **digoxin** (or related digitalis glycosides). Esculin acts as an antioxidant and anti-inflammatory agent that improves ocular microcirculation and reduces capillary permeability, thereby stabilizing the tear film. Digoxin acts as a sodium/potassium-transporting ATPase inhibitor; when applied topically, it is believed to enhance the accommodative capacity of the ciliary muscle, relieving symptoms of eye strain associated with prolonged screen use (VDT-induced asthenopia). The drug is widely utilized in China and is being investigated by institutions such as Beijing Tongren Hospital for its efficacy in pediatric and adult populations with ocular surface disorders.
