Overview
Esomeprazole + domperidone is a fixed-dose combination medication primarily used to treat gastroesophageal reflux disease (GERD), functional dyspepsia, and peptic ulcers. It combines **esomeprazole**, a proton pump inhibitor (PPI) that is the S-isomer of omeprazole, with **domperidone**, a peripheral dopamine D2 and D3 receptor antagonist. Esomeprazole acts by irreversibly inhibiting the H+/K+-ATPase enzyme system in gastric parietal cells, thereby reducing gastric acid secretion. Domperidone serves as a prokinetic agent, increasing lower esophageal sphincter tone and enhancing gastric motility and emptying, which helps prevent the reflux of stomach contents. This dual mechanism addressing both acid production and gastric motility makes it particularly effective for patients whose symptoms are not controlled by acid suppression alone.
