Drug intelligence / Profile preview
This drug regimen consists of the sequential administration of **esomeprazole**, a proton pump inhibitor (PPI), and **keverprazan**, a potassium-competitive acid blocker (P-CAB). It is being evaluated in a multicenter randomized controlled trial led by the **General Hospital of Northern Theater Command** (formerly General Hospital of Shenyang Military Region) to assess its efficacy in improving short-term outcomes following prophylactic endoscopic variceal treatment (EVT) in patients with liver cirrhosis and esophageal or gastric varices. Esomeprazole acts by irreversibly binding to the gastric H+/K+-ATPase, while keverprazan provides reversible, potassium-competitive inhibition of the same enzyme. The regimen typically involves intravenous esomeprazole (40 mg/d) for 3 to 7 days, followed by oral keverprazan (20 mg/d) for a total duration of two weeks. The primary goal is to maintain a high gastric pH to promote mucosal healing and reduce the risk of post-banding ulcer bleeding.
