**Esomeprazole + rofecoxib** is a combination of a proton pump inhibitor (*esomeprazole*) and a cyclo-oxygenase-2 (COX-2) selective nonsteroidal anti-inflammatory drug (*rofecoxib*). Esomeprazole acts by inhibiting gastric H+/K+ ATPase, thus reducing stomach acid production, while rofecoxib inhibits COX-2, reducing the formation of pro-inflammatory prostaglandins. This combination may be considered in patients needing anti-inflammatory therapy while also requiring gastric acid suppression, for example, those at risk of NSAID-induced gastrointestinal complications[1][2]. According to clinical pharmacology studies, esomeprazole does not significantly affect the pharmacokinetics of rofecoxib when administered together[1].
