Estradiol + tibolone is a hypothetical or investigational combination of two hormonal agents used in the management of menopausal symptoms. Estradiol is a natural estrogen hormone, while tibolone is a synthetic steroid with estrogenic, progestogenic, and androgenic activity. Both are used individually for hormone replacement therapy (HRT) in postmenopausal women to alleviate vasomotor symptoms (such as hot flushes), improve mood and sexual function, and prevent osteoporosis. Tibolone acts as an agonist at estrogen receptors (with preference for ER alpha), progesterone receptors, and androgen receptors through its active metabolites. While both drugs are established HRT options on their own, there is no evidence that they are marketed or clinically used together as a fixed-dose combination; rather, they may be compared head-to-head or against other HRT regimens in clinical studies[1][2][3][4][5].
