estramustine + docetaxel + carboplatin

A multi-agent chemotherapy regimen combining the small molecules estramustine, docetaxel, and carboplatin, used investigationally for castration-resistant/hormone-refractory prostate cancer. Estramustine is an estradiol–nitrogen mustard conjugate that disrupts microtubule dynamics and has antiandrogenic/estrogenic effects; docetaxel is a taxane that stabilizes microtubules and inhibits depolymerization; carboplatin is a platinum compound causing DNA crosslinks and apoptosis. The combination (often termed DEC) has shown PSA declines and objective responses with acceptable toxicity when supported with G-CSF in phase II settings for hormone-refractory prostate carcinoma, with reported median time to progression around 8–12 months and overall survival around 19–26 months in earlier-line studies, and shorter outcomes in later-line use.[1][2][5]
