This is a combination therapy consisting of three drugs—etanercept, leflunomide, and methylprednisolone—used primarily in the management of autoimmune and inflammatory conditions such as rheumatoid arthritis. Etanercept is a biologic agent that acts as a tumor necrosis factor (TNF) inhibitor, blocking the activity of TNF-alpha to reduce inflammation. Leflunomide is a disease-modifying antirheumatic drug (DMARD) that inhibits pyrimidine synthesis by blocking dihydroorotate dehydrogenase, leading to reduced lymphocyte proliferation. Methylprednisolone is a synthetic glucocorticoid corticosteroid with potent anti-inflammatory and immunosuppressive effects through modulation of gene expression and suppression of immune cell activity. This combination may be used when monotherapy or dual therapy does not provide adequate disease control[1][5][6]. Each component targets different aspects of the immune response for synergistic effect but increases risk for immunosuppression-related adverse events.
