Overview

This is a combination therapy consisting of three drugs—etanercept, methotrexate, and prednisolone—used primarily in the management of autoimmune and inflammatory diseases such as rheumatoid arthritis. - **Etanercept** is a biologic fusion protein that acts as a tumor necrosis factor (TNF) inhibitor by binding to TNF-alpha, thereby blocking its pro-inflammatory activity. It is indicated for moderate to severe rheumatoid arthritis and other inflammatory conditions[1][3][6]. - **Methotrexate** is a conventional disease-modifying antirheumatic drug (DMARD) that inhibits dihydrofolate reductase, interfering with folic acid metabolism and suppressing immune cell proliferation. Its anti-inflammatory effects are also attributed to increased adenosine signaling[4]. - **Prednisolone** is a synthetic glucocorticoid corticosteroid that exerts broad anti-inflammatory and immunosuppressive effects by modulating gene expression through the glucocorticoid receptor. The combination of etanercept with methotrexate has been shown to be more effective than either agent alone in certain populations with rheumatoid arthritis; however, the addition of prednisolone provides rapid symptomatic relief due to its potent anti-inflammatory action. This triple regimen may be used in cases where disease activity remains high despite standard therapies or during acute flares.
