Etentamig + daratumumab is a combination therapy under clinical investigation primarily for the treatment of relapsed or refractory multiple myeloma. Etentamig (ABBV-383) is a bispecific monoclonal antibody that targets B-cell maturation antigen (BCMA) and CD3, facilitating immune-mediated cytotoxicity by redirecting T-cells against BCMA-expressing multiple myeloma cells. Daratumumab is a human IgG1k monoclonal antibody targeting CD38, leading to direct tumor cell killing and immune-mediated mechanisms such as antibody-dependent cellular cytotoxicity, complement-dependent cytotoxicity, and apoptosis. This combination aims to increase anti-myeloma activity in patients who have received prior lines of therapy[7][9][5][1]. Early phase clinical trials have shown high overall response rates and manageable safety profiles in heavily pretreated populations[7][9].
