Stediril 30 is a combined oral contraceptive (COC) containing the synthetic estrogen ethinyl estradiol (0.03 mg) and the synthetic progestin levonorgestrel (0.15 mg). It functions primarily by suppressing the secretion of gonadotropins (follicle-stimulating hormone and luteinizing hormone) via the hypothalamic-pituitary-gonadal axis, thereby inhibiting the LH surge and preventing ovulation. Additionally, it alters the cervical mucus to impede sperm penetration and modifies the endometrium to reduce the likelihood of implantation. Beyond its primary use for oral contraception, Stediril 30 is utilized in managing gynecological conditions such as endometriosis and endometrial hyperplasia. It is currently being studied by Erasmus MC in Phase 4 trials to evaluate the impact of hormonal contraceptives on the hypothalamic-pituitary-adrenal (HPA) axis, specifically regarding basal cortisol levels and responses to ACTH stimulation.
