Overview
Etigilimab + nivolumab is a combination therapy composed of two monoclonal antibodies: etigilimab (an IgG1 antibody targeting TIGIT) and nivolumab (an IgG4 antibody targeting PD-1). Etigilimab inhibits TIGIT, a co-inhibitory receptor on T cells and natural killer cells, thereby enhancing antitumor immunity through blockade of TIGIT-PVR interaction, reduction of regulatory T cells, and promotion of myeloid cell activation. Nivolumab blocks PD-1-mediated immune checkpoint signaling, enabling T cells to recognize and kill tumor cells. This combination is under investigation for various advanced or metastatic solid tumors, aiming to improve efficacy especially in checkpoint-inhibitor-naïve or PD-L1-low cancers.
