Etonogestrel + testosterone undecanoate is an investigational combination of a progestin (etonogestrel) and an androgen (testosterone undecanoate) studied for use as a male hormonal contraceptive. Etonogestrel is a long-acting synthetic progestin commonly used in contraceptive implants, while testosterone undecanoate is an esterified form of testosterone used for hormone replacement therapy in hypogonadal men. The combination works by suppressing spermatogenesis through negative feedback on the hypothalamic-pituitary-gonadal axis, reducing endogenous gonadotropin secretion and thus sperm production. Clinical studies have shown that this regimen can induce rapid and consistent suppression of sperm counts to levels compatible with effective contraception[8][4][2]. This approach aims to provide reversible, reliable male contraception.
