Overview

Etoposide + cisplatin is a combination chemotherapy regimen commonly referred to as EP or PE. It is primarily used as a first-line treatment for several types of cancer, most notably small cell lung cancer (SCLC) and testicular cancer. Etoposide acts as a topoisomerase II inhibitor, interfering with DNA synthesis by causing DNA strand breaks during the S and G2 phases of the cell cycle. Cisplatin is a platinum-based compound that covalently binds to DNA, forming intra-strand and inter-strand cross-links that disrupt DNA function, ultimately inhibiting DNA, RNA, and protein synthesis in a cell cycle–nonspecific manner[2][3][5][6]. The combination exploits synergistic cytotoxic effects on rapidly dividing tumor cells.
