Etoposide + pyrotinib is a combination therapy consisting of two small molecule drugs used in the treatment of HER2-positive metastatic breast cancer (MBC), particularly in patients who have been heavily pretreated with other regimens. Pyrotinib is an irreversible pan-ErbB receptor tyrosine kinase inhibitor that targets HER1 (EGFR), HER2, and HER4, thereby blocking downstream signaling pathways involved in tumor cell proliferation and survival. Etoposide is a topoisomerase II inhibitor that interferes with DNA replication and transcription, leading to apoptosis of cancer cells. The combination has been studied using metronomic dosing (low continuous oral dosing) for both agents to maximize efficacy while minimizing toxicity. Clinical studies have shown promising results for this regimen in patients who have exhausted multiple prior lines of anti-HER2 therapy, offering an alternative when other options such as antibody-drug conjugates are not accessible or affordable[1][2][3].
