Overview

Etoricoxib + tramadol is a fixed-dose combination of the selective cyclooxygenase-2 (COX-2) inhibitor etoricoxib and the centrally acting analgesic tramadol. This combination is designed for the management of moderate to severe acute pain, such as postoperative pain following dental or orthopedic surgery. Etoricoxib provides anti-inflammatory and analgesic effects by inhibiting prostaglandin synthesis, while tramadol acts through a dual mechanism: as a mu-opioid receptor agonist and as an inhibitor of the reuptake of serotonin and norepinephrine. By targeting both peripheral and central pain pathways, the combination aims to achieve superior efficacy with potentially lower doses of each component, thereby improving the safety profile. It is primarily being developed and studied by pharmaceutical companies in Mexico, such as Laboratorios Liomont and Laboratorios Silanes.
