Overview

This is a **combination therapy consisting of ETX2514 (durlobactam), sulbactam, imipenem, and cilastatin**, investigated for the treatment of severe infections caused by carbapenem-resistant Acinetobacter baumannii-calcoaceticus complex, including hospital-acquired or ventilator-associated bacterial pneumonia, and bloodstream infections[1][7][8][10]. - **ETX2514 (durlobactam)** is a novel, diazabicyclooctenone non-beta-lactam, broad-spectrum β-lactamase inhibitor, particularly targeting class A, C, and D β-lactamases, restoring susceptibility of Acinetobacter baumannii to sulbactam and imipenem[2][4][5][6][7][9]. - **Sulbactam** is a β-lactam antibiotic and β-lactamase inhibitor that also directly targets penicillin-binding protein 3, thereby inhibiting cell wall synthesis in susceptible bacteria[5][7]. - **Imipenem** is a broad-spectrum carbapenem antibiotic targeting penicillin-binding proteins, thus inhibiting bacterial cell wall synthesis; **cilastatin** is included to prevent renal degradation of imipenem by inhibiting the enzyme dehydropeptidase I. This combination is given intravenously and has been evaluated in clinical trials (including NCT02971423, NCT03445195, and NCT03894046)[1][4][8][10]. Primary indication: serious infections due to multi-drug resistant Acinetobacter species, especially carbapenem-resistant strains.
