EV20 + Omomyc (also written as EV20/Omomyc) is a novel dual-targeting immunoconjugate in preclinical development by MediaPharma S.r.l. in collaboration with the University of Chieti-Pescara. The therapeutic is constructed by linking EV20, a humanized monoclonal antibody targeting the Human Epidermal Growth Factor Receptor 3 (HER3/ErbB-3), to Omomyc, a tissue-penetrating peptide that acts as a dominant-negative inhibitor of the MYC oncogene. The rationale behind this conjugate is to leverage the HER3-targeting specificity of EV20 to deliver the Omomyc payload directly into HER3-expressing tumor cells. Once internalized, Omomyc disrupts the formation of MYC-MAX heterodimers, thereby inhibiting the transcription of MYC-target genes essential for tumor growth and survival. This approach aims to provide a targeted delivery system for MYC inhibition, a pathway that has historically been difficult to target with traditional small molecules.
